=== Atrial myxoma === Myxomas are usually located in either the left or right atrium of the heart; about 86 percent occur in the left atrium. Myxomas are typically pedunculated, with a stalk that is attached to the interatrial septum. The most common location for attachment of the stalk is the fossa ovalis region of the interatrial septum. An atrial myxoma may create an extra heart sound, audible to auscultation just after S2. It is most seen on echocardiography, as a pedunculated mass that is heterogeneous in appearance. A left atrial myxoma will cause an increase in pulmonary capillary wedge pressure. The differential diagnosis include other cardiac tumors such as lipomas and rhabdomyomas (and rarely teratomas). These other tumors of the heart are typically not pedunculated, however, and are more likely to infiltrate the muscle of the heart. Cardiac magnetic resonance imaging (MRI) can help non-invasively diagnose cardiac tumors. However, diagnosis usually requires examination of a tissue sample by a pathologist.
A property of DNA is its ability to re-anneal into double-stranded molecules when pH conditions are neutralized. Under neutral conditions, hydrogen bonds reform between complementary base pairs. Potassium acetate reacts with the SDS detergent, magnesium ions, and calcium ions already present in the solution and forms potassium dodecyl sulfate (KDS), an insoluble white solid which precipitates out of solution. The remaining chromosomal DNA strands, denatured proteins, and added chemicals stick together and precipitate out with the KDS. The plasmid DNA, however, remains dissolved in the liquid solution. The solution is usually centrifuged to collect the insoluble precipitates into a debris pellet at the bottom of the sample tube and thereby isolate them from the supernatant.

This one-pot protocol can assemble up to 5 different fragments accurately, while several commercial providers have kits to accurately assemble up to 15 different fragments in a two-step reaction. This reliance on PCR may also affect the fidelity of the reaction when long fragments, fragments with high GC content or repeat sequences are used.

== Tetramers in immunology == In immunology, MHC tetramers can be used in tetramer assays, to quantify numbers of antigen-specific T cells (especially CD8+ T cells). MHC tetramers are based on recombinant class I molecules that, through the action of bacterial BirA, have been biotinylated. These molecules are folded with the peptide of interest and β2M and tetramerized by a fluorescently labeled streptavidin. (Streptavidin binds to four biotins per molecule.) This tetramer reagent will specifically label T cells that express T cell receptors that are specific for a given peptide-MHC complex. For example, a Kb/FAPGNYPAL tetramer will specifically bind to Sendai virus specific cytotoxic T cell in a C57BL/6 mouse. Antigen specific responses can be measured as CD8+, tetramer+ T cells as a fraction of all CD8+ lymphocytes. The reason for using a tetramer, as opposed to a single labeled MHC class I molecule is that the tetrahedral tetramers can bind to three TCRs at once, allowing specific binding in spite of the low (1 micromolar) affinity of the typical class I-peptide-TCR interaction. MHC class II tetramers can also be made, although these are more difficult to work with practically.
