== Metabolism == The two bioactive forms of vitamin B12 are methylcobalamin in cytosol and adenosylcobalamin in mitochondria. The cyanide is converted to thiocyanate and excreted by the kidney. The MMACHC gene product catalyzes the decyanation of cyanocobalamin as well as the dealkylation of alkylcobalamins including methylcobalamin and adenosylcobalamin. The MMACHC gene product is characterised as a cyanocobalamin reductase (cyanide-eliminating) and an alkylcobalamin reductase. It enables the interconversion of cyano- and alkylcobalamins. Endogenous vitamin B12 becomes inactive when exposed to hydrogen cyanide and nitric oxide in cigarette smoke. More specifically, it causes the active forms to convert to cyanocobalamin, causing a functional deficiency. Functional vitamin B12 deficiency can also develop with heavy regular use of nitrous oxide N2O, also known as "laughing gas", by oxidizing the cobalt atom and inactivating the vitamin. Used for anaesthesia in a clinical setting or as a propellant gas, it's commonly abused as a recreational drug. The Clearwater-based company retained control over restaurants in the Tampa Bay Area, Chicago metropolitan area, and one in Manhattan, as well as rights to develop a Hooters casino and sell sauces in grocery stores, while all other locations were under the aegis of Hooters of America, which sold franchising rights to the rest of the United States and international locations. == Research == The combination drug dextromethorphan/quinidine (AVP-923), traditionally used to treat pseudobulbar affect, is under investigation for the treatment of a variety of other neurological and neuropsychiatric conditions including agitation associated with Alzheimer's disease, among others. In 2013, a randomized clinical trial found that dextromethorphan may reduce the overall discomfort and duration of withdrawal symptoms associated with opioid use disorder. When combined with clonidine, dextromethorphan reduced the overall time needed for withdrawal symptoms to peak by 24 hours while reducing severity of symptoms compared to clonidine alone.
=== Stimulants === Stimulants are drugs that usually act on the central nervous system to modulate mental function and behavior, increasing an individual's sense of excitement, decreasing the sensation of fatigue and improving motor coordination. The latter happens via improvement of the interaction between nervous system and musculature. It has greater effect when an athlete is already exhausted - the period when the coordination suffers the most. In the World Anti-Doping Agency list of prohibited substances, stimulants are the second largest class after the anabolic steroids. Benzedrine is a trade name for amphetamine. The Council of Europe says it first appeared in sport at the Berlin Olympics in 1936. It was produced in 1887 and the derivative, Benzedrine, was isolated in the U.S. in 1934 by Gordon Alles. Its perceived effects gave it the street name "speed". British troops used 72 million amphetamine tablets in the Second World War and the RAF got through so many that "Methedrine won the Battle of Britain" according to one report. The problem was that amphetamine leads to a lack of judgement and a willingness to take risks, which in sport could lead to better performances but in fighters and bombers led to more crash landings than the RAF could tolerate. The drug was withdrawn but large stocks remained on the black market. Amphetamine was also used legally as an aid to slimming and also as a thymoleptic before being phased out by the appearance of newer agents in the 1950s.

=== Luminous flux: watt equivalent === With the phaseout of the incandescent lamp in the United States and European Union in the early 21st century, manufacturers and sellers of more energy-efficient lamps have compared the visible light output of their lamps to commonly used incandescent lamp sizes with the watt equivalent or watt incandescent replacement (usually with a lowercase w as a unit symbol, as opposed to capital W for the actual wattage). 1 watt incandescent replacement corresponds to 15 lumens. Thus, a 72-watt halogen lamp, a 23-watt compact fluorescent lamp and a 14-watt light-emitting diode lamp, all of which emit 1500 lumens of visible light, are all marketed as "100 watt incandescent replacement" (100w).
If the target DNA contains an allele complementary to the probe's 3’ base, the target DNA will completely hybridize to the probe, allowing DNA polymerase to extend from the 3’ end of the probe. This is detected by the incorporation of the fluorescently labeled dNTPs onto the end of the probe. If the target DNA does not contain an allele complementary to the probe's 3’ base, the target DNA will produce a mismatch at the 3’ end of the probe and DNA polymerase will not be able to extend from the 3' end of the probe. The benefit of the second approach is that several labeled dNTPs may get incorporated into the growing strand, allowing for increased signal. However, DNA polymerase in some rare cases, can extend from mismatched 3’ probes giving a false positive result. A different approach is used by Sequenom's iPLEX SNP genotyping method, which uses a MassARRAY mass spectrometer. Extension probes are designed in such a way that 40 different SNP assays can be amplified and analyzed in a PCR cocktail. === Pancreas === The human fetal pancreas begins to develop by the fourth week of gestation. Five weeks later, the pancreatic alpha and beta cells have begun to emerge. Reaching eight to ten weeks into development, the pancreas starts producing insulin, glucagon, somatostatin, and pancreatic polypeptide. During the early stages of fetal development, the number of pancreatic alpha cells outnumbers the number of pancreatic beta cells. The alpha cells reach their peak in the middle stage of gestation. From the middle stage until term, the beta cells continue to increase in number until they reach an approximate 1:1 ratio with the alpha cells. The insulin concentration within the fetal pancreas is 3.6 pmol/g at seven to ten weeks, which rises to 30 pmol/g at 16–25 weeks of gestation. Near term, the insulin concentration increases to 93 pmol/g. The endocrine cells have dispersed throughout the body within 10 weeks. At 31 weeks of development, the islets of Langerhans have differentiated. While the fetal pancreas has functional beta cells by 14 to 24 weeks of gestation, the amount of insulin that is released into the bloodstream is relatively low. In a study of pregnant women carrying fetuses in the mid-gestation and near term stages of development, the fetuses did not have an increase in plasma insulin levels in response to injections of high levels of glucose.
