This is a working overview of 非天然氨基酸, written for readers who want more than a one-paragraph summary but less than a textbook.

Signal transduction begins when the peptide binds GHSR-1a on pituitary somatotrophs. The receptor couples to Gq/11 proteins, activating phospholipase C, which cleaves phosphatidylinositol bisphosphate into inositol trisphosphate and diacylglycerol. Inositol trisphosphate releases calcium from intracellular stores, and the resulting rise in cytosolic calcium drives growth hormone vesicle fusion. Concurrent Gs coupling and cyclic AMP elevation have also been reported, and the relative contribution of each arm to the overall secretory response is not fully settled.
Structural features distinguish the molecule from earlier secretagogues. An alpha-aminoisobutyric acid residue near the N-terminus and a D-naphthylalanine substitution increase receptor affinity, while C-terminal amidation improves resistance to exopeptidases. These modifications are associated with reduced stimulation of appetite and of the hypothalamic-pituitary-adrenal axis compared with hexarelin or growth hormone releasing peptide-6. Whether the same profile applies at every dose level studied is a matter of ongoing investigation rather than settled consensus.

Compared with earlier growth hormone secretagogues such as GHRP-6 and hexarelin, ipamorelin has been reported to produce less stimulation of adrenocorticotropic hormone, cortisol, and prolactin in animal and early human studies. This selectivity is usually attributed to differences in receptor subtype interactions and to the tissue distribution of the receptor. Effects on appetite appear weaker than those of ghrelin itself, although the supporting evidence base is small. Whether these differences produce a distinct clinical profile remains an open question, since controlled human trials are limited.
Ipamorelin is a synthetic pentapeptide first described in the 1990s by researchers at Novo Nordisk during a program to develop selective growth hormone secretagogues. Its sequence is Aib-His-D-2-Nal-D-Phe-Lys-NH2, incorporating two non-natural residues, alpha-aminoisobutyric acid and D-2-naphthylalanine. The C-terminus is amidated, and the material is supplied as a white lyophilized powder. The molecular formula is C38H49N9O5 and the monoisotopic mass is approximately 711.85 daltons. The short chain and modified residues give it greater resistance to enzymatic degradation than many larger peptide hormones.
Compared with other secretagogues such as GHRP-2, GHRP-6, and hexarelin, ipamorelin is described as more selective. Published animal work reports little or no increase in adrenocorticotropic hormone, cortisol, or prolactin at doses that release growth hormone. This selectivity is the property most often cited in the research literature. Whether the same profile holds across species and dosing schedules remains an open question, since human data are limited and come largely from small studies.
Ipamorelin is a synthetic pentapeptide that belongs to the growth hormone secretagogue family. Its sequence is Aib-His-D-2-Nal-D-Phe-Lys-NH2, and its molecular mass is approximately 711.9 daltons. The compound was described in the late 1990s by researchers seeking molecules that release growth hormone with fewer side effects than earlier secretagogues. It is a laboratory and research compound, not an approved medicine in most jurisdictions.
At the molecular level, ipamorelin acts as an agonist at the growth hormone secretagogue receptor, also called the ghrelin receptor or GHS-R1a. Binding to this receptor on pituitary somatotroph cells triggers a signaling cascade that leads to growth hormone release. The effect is mediated through phospholipase C and calcium mobilization rather than through the cyclic AMP pathway used by growth hormone releasing hormone. The two pathways are complementary, and combined stimulation produces a larger response than either alone.
The therapeutic uses of puffer fish (tetraodon) eggs were mentioned in the first Chinese pharmacopoeia Pen-T'so Ching (The Book of Herbs, allegedly 2838–2698 BC by Shennong; but a later date is more likely), where they were classified as having "medium" toxicity, but could have a tonic effect when used at the correct dose. The principal use was "to arrest convulsive diseases". In the Pen-T'so Kang Mu (Index Herbacea or The Great Herbal by Li Shih-Chen, 1596) some types of the fish Ho-Tun (the current Chinese name for tetraodon) were also recognized as both toxic yet, at the right dose, useful as part of a tonic. Increased toxicity in Ho-Tun was noted in fish caught at sea (rather than river) after the month of March. It was recognized that the most poisonous parts were the liver and eggs, but that toxicity could be reduced by soaking the eggs. (Tetrodotoxin is slightly water-soluble, and soluble at 1 mg/ml in slightly acidic solutions.) The German physician Engelbert Kaempfer, in his "A History of Japan" (translated and published in English in 1727), described how well known the toxic effects of the fish were, to the extent that it would be used for suicide and that the Emperor specifically decreed that soldiers were not permitted to eat it. There is also evidence from other sources that knowledge of such toxicity was widespread throughout southeast Asia and India. The first recorded cases of TTX poisoning affecting Westerners are from the logs of Captain James Cook from 7 September 1774. The crew received a fish from the New Caledonia natives.
=== Boiling points === Carboxylic acids tend to have higher boiling points than water, because of their greater surface areas and their tendency to form stabilized dimers through hydrogen bonds. For boiling to occur, either the dimer bonds must be broken or the entire dimer arrangement must be vaporized, increasing the enthalpy of vaporization requirements significantly.
[T]he Iraqi people, who were not consulted about the invasion, have paid the price for their government's madness ... Iraqis understood the legitimacy of a military action to drive their army from Kuwait, but they have had difficulty comprehending the Allied rationale for using air power to systematically destroy or cripple Iraqi infrastructure and industry: electric power stations (92 percent of installed capacity destroyed), refineries (80 percent of production capacity), petrochemical complexes, telecommunications centers (including 135 telephone networks), bridges (more than 100), roads, highways, railroads, hundreds of locomotives and boxcars full of goods, radio and television broadcasting stations, cement plants, and factories producing aluminum, textiles, electric cables, and medical supplies. However, the UN subsequently spent billions rebuilding hospitals, schools, and water purification facilities throughout the country.
==== Detection ==== Cabbage loopers possess olfactory receptor neurons on their antennae for detecting pheromones. The neurons are specifically located on two sensory structures called sensilla that differ in length and pore density. Male loopers have two types of neurons, and depending on which sensilla that are present, the neurons will detect female pheromones at varying sensitivities to each of the six pheromones. The neurons are most sensitive to the main component of the female pheromone blend, cis-7-dodecenyl acetate, and the male inhibitory signal, cis-7-dodecenol. The presence of cis-7-dodecenyl acetate is crucial for male response to female pheromones, as it is 80% of the entire blend. The base region of the antennae, where receptor neurons for this pheromone are located, has more sensory structures than the ends. The base region is also less likely to experience damage, showing the importance of detecting the pheromone. It is not clear why male neurons detect the inhibitory compound, as there is no evidence showing that females produce this compound. One possibility is that its presence in the female pheromone blend may be too small to be detected by scientific equipment. The inhibitory signal only elicits a response when delivered alongside female pheromones to avoid mixing signals from other species, suggesting that while it cannot be detected in the female pheromone blend, it has an important role in female detection. These neurons are also capable of recognizing and responding to cis-7-tetradecenyl acetate and cis-9-tetradecenyl acetate.
According to Future Forward PAC, a derivative of his campaign's "Kamala is for they/them, President Trump is for you" attack ad (with commentary by Charlamagne tha God spliced in) was one of the most effective of the campaign, shifting the race 2.7 percentage points in favor of Trump after viewers watched it. However, findings from an RCT study released by Ground Media/GLAAD said that the ad did not have an impact on who viewers intended to vote for.

Nanoparticles are classified as having at least one of its dimensions in the range of 1-100 nanometers (nm). The small size of nanoparticles allows them to have unique characteristics which may not be possible on the macro-scale. Self-assembly is the spontaneous organization of smaller subunits to form larger, well-organized patterns. For nanoparticles, this spontaneous assembly is a consequence of interactions between the particles aimed at achieving a thermodynamic equilibrium and reducing the system's free energy. The thermodynamics definition of self-assembly was introduced by Professor Nicholas A. Kotov. He describes self-assembly as a process where components of the system acquire non-random spatial distribution with respect to each other and the boundaries of the system. This definition allows one to account for mass and energy fluxes taking place in the self-assembly processes. This process occurs at all size scales, in the form of either static or dynamic self-assembly. Static self-assembly utilizes interactions amongst the nano-particles to achieve a free-energy minimum. In solutions, it is an outcome of random motion of particles and the affinity of their binding sites for one another. A dynamic system is forced to not reach equilibrium by supplying the system with a continuous, external source of energy to balance attractive and repulsive forces. Magnetic fields, electric fields, ultrasound fields, light fields, etc. have all been used as external energy sources to program robot swarms at small scales.

=== Controlling cell volume === Failure of the Na+-K+ pumps can result in swelling of the cell. A cell's osmotic concentration is the sum of the concentrations of the various ion species and many proteins and other organic compounds inside the cell. When this is higher than the osmotic concentration outside of the cell, water flows into the cell through osmosis. This will cause the cell to swell up and lyse. The Na+-K+ pump helps to maintain the right concentrations of ions. Furthermore, when the cell begins to swell, this automatically activates the Na+-K+ pump because it changes the internal concentrations of Na+-K+ to which the pump is sensitive.
== Diagnosis == Echocardiography is the preferred screening test in the diagnosis of PAH as it accurately estimates pulmonary pressures. Other causes of pulmonary hypertension, such as left heart disease (WHO group 2), pulmonary hypertension due to lung disease (such as COPD)(WHO group 3) and pulmonary hypertension due to chronic blood clots in the pulmonary arteries (WHO group 4) need to be ruled out. Some tests which are commonly used include a CT scan of the chest, ventilation perfusion scan or a CT angiography (to rule out pulmonary hypertension due to chronic blood clots), pulmonary function test, and a cardiac MRI. Cardiac MRI is the preferred test to evaluate right ventricular structure and function in PAH. Pulmonary function testing in PAH may show an obstructive or restrictive defect, and the diffusion capacity of carbon monoxide (used as a surrogate for gas exchange in the alveoli) is reduced. A sleep study helps to rule out sleep disordered breathing, such as sleep apnea, which may contribute to PAH. The N-terminal prohormone of brain natriuretic peptide (NT pro-BNP) may be monitored in those with PAH and is prognostic. A right heart catheterization to measure right heart pressure is required for the diagnosis of PAH.
=== Wild birds and fish === Thiamine deficiency has been identified as the cause of a paralytic disease affecting wild birds in the Baltic Sea area dating back to 1982. In this condition, there is difficulty in keeping the wings folded along the side of the body when resting, loss of the ability to fly and voice, with eventual paralysis of the wings and legs and death. It affects primarily 0.5–1 kg-sized birds such as the European herring gull (Larus argentatus), common starling (Sturnus vulgaris), and common eider (Somateria mollissima). Researchers noted, "Because the investigated species occupy a wide range of ecological niches and positions in the food web, we are open to the possibility that other animal classes may develop thiamine deficiency, as well."p. 12006 In the counties of Blekinge and Skåne, mass deaths of several bird species, especially the European herring gull, have been observed since the early 2000s. More recently, species of other classes seems to be affected. High mortality of salmon (Salmo salar) in the river Mörrumsån is reported, and mammals such as the Eurasian elk (Alces alces) have died in unusually high numbers. Lack of thiamine is the common denominator where analysis is done. In April 2012, the County Administrative Board of Blekinge found the situation so alarming that they asked the Swedish government to set up a closer investigation.

Injury in humans has been studied extensively for its importance in medicine. Much of medical practice, including emergency medicine and pain management, is dedicated to treating injuries. The World Health Organization has developed a classification of injuries in humans by categories, including mechanism, objects/substances producing injury, place of occurrence, activity when injured, and the role of human intent. In addition to physical harm, injuries can cause psychological harm, including post-traumatic stress disorder, or depression (mood).

Both molecules activate the same receptor, GHSR-1a, but they share little sequence identity. Ghrelin is a 28-amino-acid hormone carrying a distinctive acyl modification, whereas ipamorelin is a short synthetic peptide. The shared target explains overlapping endocrine effects, while the different structures account for differences in metabolic stability and receptor selectivity.
Early animal and human studies reported growth hormone release at levels that produced small changes in cortisol, prolactin and adrenocorticotropic hormone. That contrast with older secretagogues gave rise to the selectivity label. The observation is consistent across several reports but rests on a limited number of small studies.
Plasma peptidases cleave the molecule into smaller fragments, and reported half-lives in human studies are on the order of a couple of hours. The identity and activity of individual breakdown products have not been fully mapped. Questions about tissue-level clearance remain open.
